METHOD :
Kinetic color test
PRINCIPLE
In acid environment, alpha-naphthyl phosphate is hydrolysed by acid
phosphatase to produce alpha-napthol and phospate. Alpha-napthol
reacts with diazo-2-chloro-5-toluene (Fast Red TR Salt) forming
an azo dye compound which absorbs maximally at 405 nm and is directly
proportional to total acid phosphatase activity. When the activity
is measured in the presence of tartarate, the prostatic activity
is inhibited. The difference between Total (ACPT) and Non-prostatic
acid phosphatase (ACPNP) corresponds to the prostatic fraction.

Reagent II : Substrate reagent
Reagent III : Tartarate reagent
Reagent IV: Acetate reagent
SAMPLE
Serum. Do not use plasma.
Use fresh serum or Acidify 1ml of serum with 20
µl Acetate reagent
& store at 2 - 8°C till further use.

REAGENT PREPARATION
(A) Total acid phosphatase:
Dissolve the contents of one vial of
Substrate with the
volume of buffer as specified on the vial & label it as 'A'.
(B) Non-prostatic acid phosphatase:
Dissolve the contents of one vial of
Substrate with the
volume of buffer as specified on the vial & label it as B,
then add 10 ul of Sodium Tartarate in 1ml of reconstituted
reagent.
STABILITY : 7 days at 2-8° C/6 hrs
at R.T.

MANUAL PROCEDURE
PIPETTE directly into tubes A or B 1 ml of reagent
Then add
Mix well and after 5 min 37°C read absorbance values and start
timer simultaneously. Measure absorbance increase every minute during
