METHOD
Kinetic colorimetric test
PRINCIPLE
2-chloro-4-nitrophenyl-alpha-D-maltotrioside (CNPG3)
is hydrolyzed by alpha-amylase to produce 2-chloro-4-nitrophenol (CNP).
The amount of 2-chloro-4-nitrophenol, monitored at 405 nm, is directly
proporational to alpha-amylase activity.
SAMPLE:
Serum, heparinized plasma urine diluted 1:2.
STABILITY
serum upto 7 days at R.T. 3 months at 2-8°C
Urine : 2 days at R.T. 10 days at 2-8°C

MANUAL PROCEDURE
PIPETTE INTO TEST TUBE

Mix well and let stand for 1 min. read initial absorbance and start
timer simultaneously. Measure absorbance increase every minute for
3 minutes (p A/min).
CALCULATION AND LINEARITY

This method is linear upto a concentration of 2000 U/l
sample above this concentration should be diluted 1:2.
Multiply the result by 2.
NOTES Avoid contamination of reagent, samples and glassware by
saliva because it has a high amylase content. Do not pipette by mouth.
Avoid Haemolysis.
Avoid anticoagulants as EDTA or sodium citrate. |
