METHOD
Colorimetric endpoint test

PRINCIPLE

Chloride reagent reacts with mercurous thiocyanate to form mercury perchlorate and thiocyanate. Thiocyanate forms a red complex with ferric ions in the presence of nitric acid.

SAMPLE
Serum or heparinised plasma.
Urine diluted 1:2 with distilled water.

NOTE:

CHLOM 50 are specially pre-treated monovials with 1 ML pre-dispensed reagent. Just add 10 µl sample / std. & aspirate. Use the same programme.

MANUAL PROCEDURE PIPETTE INTO TEST TUBE

Mix well, incubate at 20 - 25°C for 5 min. Measure final absorbance of the sample (Ac) and standard (As) against the reagent blank.

CALCULATION AND LINEARITY
Ac/As x C = mEq/l SERUM or PLASMA
Ac/Ac x C x 2 = mEq/l URINE

C = Concentration Standard .

The method is linear to a concentration of 130 mEq/l.
