METHOD
 UV endpoint test

PRINCIPLE
 Inorganic phosphate reacts in acid environment with molybdic acid to form an unreduced phosphomolybdic acid complex, which absorbs light at 340 nm. The absorbance is directly proportional to the phosphorus concentration in the sample.

SAMPLE:
Serum. Plasma, 24h Urine diluted 1:20 with distilled water.

REFERENCE VALUES
SERUM  
Men 2.1 - 5.6 mg/dl
Women 1.5 - 6.8 mg/dl
Children 4.0 - 7.0 mg/dl
24 h Urine 0.3 - 1.0 mg/dl

The reference values are only indicative in nature. Every laboratory should establish its own normal ranges.

MANUAL PROCEDURE

Mix well and incubate at RT for 10 minutes. Measure final absorbance
of the sample (Ac) and standard (As) against the reagent blank.

CALCULATION AND LINEARITY
Ac / As x Conc. Std = mg / dl Serum
Ac / As x Conc. Std x 20 = mg / dl urine

Where Vdl is the urine volume in dl
The method is linear to a concentration of 12.0 mg/dl.
