METHOD
 Colorimetric endpoint test


PRINCIPLE
Cupric ions, in an alkaline medium, interact with protein peptide bonds resulting in the formation of a coloured complex.


REAGENTS
 Reagent I : Biuret Reagent
Protein Standrad: 6 g/ dl (store at 2-8°C)


SAMPLE:
Serum. Heparinised plasma or EDTA plasma.

MANUAL PROCEDURE

 PIPETTE INTO TEST TUBE
  BLANK STD SAMPLE
SAMPLE - - 20 µl
STANDARD - 20 µl -
REAGENT 1000 µl 1000 µl 1000 µl

Mix well, incubate for 10 minutes at 20-25°C.
Measure absorbance of the sample (Ac) and
standard (As) against reagent blank.

CALCULATION AND LINEARITY
Ac / As X conc. Std. = g/dl PROTEIN

The method is linear to a concentration of 10 g/dl.
If the concentration of the sample exceeds 10 g/dl,
dilute 1+1 with 0.9% saline solution. Multily the result by 2.
